Bacterial Alkaline Protease/Alpha-Amylase for Granular Detergent Applications

Description

Deterzyme PAG 520/220 is a blend of two active enzyme preparations as described below.

Protease: A bacterial alkaline protease (E.C.3.4.21.14), produced by the controlled fermentation of Bacillus licheniformis which is an endopeptidase capable of hydrolyzing the interior peptide bonds of protein molecules. The broad substrate specificity enables the enzyme to effectively hydrolyze most proteins. Hemoglobin, casein, e.g. yolk, soya, gelatin, fish and other proteins are hydrolyzed to lower molecular weight peptides.

Amylase: A bacterial alpha-amylase (3-2. 1, 1) of Bacillus licheniformis var. origin which is an endoamylase capable of randomly hydrolyzing the alpha-1, 4-glucosidic linkages of starch. The enzyme hydrolyzes starch, amylose and amylopectin to soluble dextrins and small quantities of glucose and maltose.

Activators and Cofactors

Deterzyme PAG 520/220 alpha-amylase is a calcium metallo-protein. However, calcium is bound very tightly by the enzyme and the need for high concentrations of calcium is eliminated. Maximum stability at extremes of pH and high temperatures is observed with 50 ppm calcium in the presence of substrate or 5000 ppm calcium in the absence of substrate.

No activators or cofactors are required for the complete activity of Deterzyme PAG 520/220.

Activity

One Detergent Alkaline Protease Unit (DAPU) is that activity which will liberate four micromoles (4 X 106 moles) of tyrosine per minute under the conditions of the assay. A copy of the Detergent Alkaline Protease Unit procedure is available upon request.

One Modified Wohlgemuth Unit (MWU) is that activity which will hydrolyze one (1) milligram of soluble starch to a defined size dextrin in thirty (30) minutes under the conditions of the assay. A copy of the Determination of Liquefying Amylase (Modified Wohlgemuth Method) is available upon request.

Effect of pH

Deterzyme PAG 520/220 has an effective pH range of pH 7 to 9 and is stable in aqueous solutions over the range of pH 6 to 10.

Effect of Temperature

In the presence of 0.60% w/v casein and 0.03% w/v STPP, Deterzyme PAG 520/220 exhibits maximum activity at 60C.The enzyme effectively hydrolyzes casein at temperatures up to 70C. Deterzyme PAG 520/220 is very stable in dilute solutions at temperatures up to 50C.

Solubility

Deterzyme PAG 520/220 is soluble in water and is compatible with most components of laundry detergents.

Use Level

Typical use levels for enzymes in detergents range from 0.5 to 3 0 percent (w/w).

Before using, the Material Safety Data Sheet for each product should be reviewed with all personnel involved for proper precautions.

Packaging

Deterzyme PAG 520/220 is available in the following standard containers: 50 Kg Polyethylene fiber drums.

Storage Conditions

In sealed containers under cool conditions Deterzyme PAG 520/220 is very stable with a loss in activity of no more than 10.0 percent in six months. Storage life can be extended by storing under refrigeration at 5C.
