Thermal Stable Bacterial Alpha Amylase
Description
Thermozyme L-340, a bacterial alpha-amylase (E.C. 3.2.1.1) produced by the controlled fermentation of Bacillus licheniformis, is an endoamylase capable of randomly hydrolyzing the interior alpha-D-(1,4) glucosidic linkages of starch. The enzyme hydrolyzes starch, amylose and amylopectin to soluble dextrins and small quantities of glucose and maltose.

Activity
Thermozyme L-340 is standardized at 340,000 MWU/ml. One Modified Wolgemuth Unit (MWU) is that activity which will hydrolyze one milligram of soluble starch to a defined size dextrin in thirty (30) minutes under the conditions of the assay. A copy of the Modified Wolgemuth procedure is available upon request.
Activators and Cofactors
Thermozyme L-340 is a calcium metalloprotein. Calcium is bound very tightly by the enzyme thus reducing the concentration of calcium required. Maximum stability at extremes of pH and temperature, with substrate present, is observed at 50 PPM calcium. In the absence of substrate, 5000 PPM provides maximum stability.
Effect of pH
Thermozyme L-340 has an optimum pH range of pH 5.5 to 7.0. The enzyme is stable in dilute solutions over the range of pH 5 to 11.

Effect of Temperature
Thermozyme L-340 has a temperature optimum over 90C at pH 6.0 in the presence of 50 PPM calcium. In the presence of substrate, the enzyme is effective at temperatures exceeding 100C.

Solubility
Thermozyme L-340 is completely soluble in water and is compatible with most laundry detergents.

Safety
Thermozyme L-340 is formulated to provide maximum safety when properly handled. In certain individuals, exposure of enzymes to skin, eyes or mucous membranes may result in irritation. The Material Safety Data Sheet should be reviewed with all personnel involved for proper precautions.
Use Level
Enzyme requirements are generally dictated by processing conditions. They will vary with substrate concentration, pH, temperature, and time. In general, increasing the Thermozyme L-340 concentration will increase the rate of starch hydrolysis and shorten the time necessary for hydrolysis. Initial evaluations of Thermozyme L-340 are made at a concentration of 0.2% to 1% of the detergent formulation. Trial applications, however, should be made to determine the final usage level.
Storage Stability
In sealed containers, at room temperature, the loss of activity is normally less than 10% in six months. Storage life can be extended by storing under refrigeration at 5C (41F).
